The stakes for Daredevil: Born Again Season 2 are extremely high after the last season's ending. Matt is surrounded on all sides by Fisk's control after he placed New York under martial law and dedicated a task force to capturing him. By the end of the first season, Matt decides to fight back against him by putting together his own team, leading to a lot of great additions in the next season.
Another aspect that stood out in the Season One finale was Heather's interaction with Fisk, where she appears to be taking a much more involved job under his mayoralship. Heather was Matt's love interest throughout the first season, and the opposite sides that they stand at by the end of it paint an ever-changing picture for Matt's love life.

Matt and Heather's future in Daredevil: Born Again Season 2

Matt woke up after the attack at the gala to ask for Karen instead of Heather. This moment led to Heather walking away from their relationship. However, the rift between them had long been forming. From her aversion to vigilantes, finding them nothing more than serial killers, to her calling Matt's warnings about Kingpin's paranoia, it was clear that there was plenty of discord between them.
Matt Murdock also accepts at one point that he feels like he's living a fake life when he's around her. A big part of that is also true, considering he can never be his real self in front of her and has to pretend to be normal. These dynamics made it clear that their relationship wasn't one to survive, but her choosing to work even more deeply with Fisk while he's trying his best to save New York sealed the deal for them.
The writer for Daredevil: Born Again, Dario Scardapane, in a recent conversation about the new season, referred to Heather as Matt's ex, even though a proper breakup was never seen in the show:
"You have two camps now. You have the resistance, and you have the administration or Fisk’s City Hall. And there are characters in both of those camps whose stories have just barely started… you have an ex of Matt, Heather, who’s now the mental health czar… So we have a lot of these characters that really are just now starting their stories in Season 2."
All of this goes to prove that with high probability, Daredevil: Born Again will not be exploring Matt's connection with Dr. Heather Glenn. We still might get to see Heather as she falls deeper into Fisk's manipulations.
What will Matt's love life look like in Peacemaker Season 2

Now that it's more than obvious that Matt and Heather's relationship is not a possibility for the second season of Daredevil: Born Again Season 2, we can consider other possibilities regarding what might happen next season.
Close to the Daredevil: Born Again Season 1, Matt notices Karen's heartbeat fastening around him, in addition to Frank's, when Karen enters. So, the second season might explore the possibility of a love triangle between the three.
The second season might even choose to explore Matt's connection with some of the new additions to Season 2, who will be joining his side of the camp.
We'll have to wait until Daredevil: Born Again Season 2 to find out how Matt's love life pans out, but whatever happens, there will for sure be a lot of action and drama from both camps to get us through.
