Penelope Milford is the daughter of Richard George Milford and Ann Marie (nee Felt) Milford. She was born in St. Louis, Missouri, but later moved to Illinois. She did not come from a large family. Milford had three siblings: Douglas Milford, Candace Saint, and her late brother, Richard Kim Milford. Not much is known about Penelope's family except for her actor brother, Kim.

Like his older sister, Kim Milford was an actor, but he was also a composer and a singer-songwriter. He was famous for his stage acting and appeared in musicals such as The Rocky Horror Show and Jesus Christ Superstar. Throughout his life, he needed surgery because of his congenital heart defects that he was born with. Kim passed away at the age of 37 after an open-heart surgery.

Milford was married to American-born poet, former actor, and activist Michael Lally for two years (1982-1984).

She is survived by her two siblings. According to The Hollywood Reporter, Penelope Milford's death was confirmed by both Douglas Milford and Candace Saint. However, the cause of death was not revealed to the public.
