In 1999, she made her TV debut with the show Early Edition, where she played the role of Susan Schwartz.

Landecker gained popularity for her role as Sarah Pfefferman in the critically acclaimed show Transparent (2014–2019). Bradley Whitford and Amy Landecker met each other for the first time in 2014 and started dating in the summer of 2015. In September 2015, the couple made their red carpet debut at the Creative Arts Emmy Awards. In a 2015 interview with People, Whitford talked about their relationship and said, “Things with Amy are spectacular. I met her last year, but we didn’t start dating for a little while. I work slowly.”

In an interview with People, Landecker also shared details about their relationship and said, “I knew Bradley for a year before we dated and I kind of thought that he seemed like a cocky actor boy and I wasn’t really that interested. In a 2020 interview with Us Weekly, Landecker talked about their relationship and family and said, “We’ve been together almost five years, but we went really slow because of the kids, the blending of the families. All of them are nice. Everyone’s well-intentioned. … I just have the greatest step-kids, and then my kid is really kind. They’re just good kids!”
