Rhimes was born in Chicago, Illinois, the youngest of six children born to university administrator Ilee Rhimes Jr. and college professor Vera P. (née Cain). Along with her three older sisters and two older brothers, Rhimes resided in Park Forest South (now University Park, Illinois). According to her, she exhibited a talent for storytelling from a young age. She developed an interest in hospital settings while volunteering in a hospital throughout her high school years.
Rhimes went to Marian Catholic High School in Chicago Heights, IL. She majored in English and cinema studies at Dartmouth College, where she graduated in 1991 with a bachelor's degree. She became a member of the Black Underground Theater Association at Dartmouth. She divided her time between writing fiction and directing and acting in student performances. She contributed to the college newspaper as well.
She relocated to San Francisco with an older sibling to work in advertising at McCann Erickson after graduating from college. She subsequently moved to Los Angeles to attend the University of Southern California to obtain a degree in screenwriting. Rhimes placed first in her USC class and was awarded the Gary Rosenberg Writing Fellowship. She earned a Master of Fine Arts degree from the USC School of Cinematic Arts.
Debra Martin Chase recruited Rhimes as an intern at USC. Rhimes owes some of her early success to mentors like well-known African-American producer Chase. Later, in The Princess Diaries 2, Chase mentored Rhimes once again. Rhimes was also hired by Mundy Lane Entertainment, which was owned by Denzel Washington.
