She started her career at the age of 10, debuting as the lead in the TV show Thunder (1977). She made her film debut in a short film, Something Queer is Happening at the Library (1978). She appeared in many TV shows and movies like Monk (2004-2009), 17 Again (2009), Transparent (2014-2019), and A Million Little Things (2019-2021). Apart from her acting in TV and films, she also did theater. She made her Broadway debut in 2008 as Roxie Hart in the famous musical Chicago.


She has played many roles, but her role as Jan remains her most loved character. Melora’s performance as Jan was highly praised because she made the character both funny and dramatic.
